Make Ink Sing is a side quest in Luciola Forest. It is unlocked after Luciola Forest main quest 2: Battle Practice. Make Ink Sing is the first quest in a 3-part quest chain.

Quest Tactics[]
This actually isn't very useful in and of itself, but it unlocks a very valuable ability eventually, so you might as well get it out of the way early. (Well, very valuable if you're attached to completing sidequests.)
The best source of Essence in Luciola Forest is Kai. If you have access to the Elder Library, Paper Wignows are a better source.
Poison can be found in Wooden Chests in Luciola Forest. The best source in Luciola Forest is Aracni. Late in the game, Oncara is also a good source.
You will later need Leafsung Ink in a few large side quests, and in crafting boots. This quest chain ends in being able to craft your own Leafsung Ink.

Kyiat approached me and asked if I wanted to learn the secret of her ink. Of course, I immediately said yes. I've seen that ink in action before, poured into a pattern that made a simple sword into one that was luckier than it had previously been. She smirked, as if she'd known what my answer was going to be all along, and snapped at me to hurry up and get some Essence and Spider Poison. I'm intrigued. Does this mean the poison itself burns the pattern into the wood? Is it the Essence? Or a reaction from mixing both?
